X-tremeGENE HP DNA Transfection Reagent is a high performance transfection reagent, free of animal-derived components. It is designed to transfect a broad range of eukaryotic cells, including insect cells, many cell lines not transfected well by other reagents, and hard-to-transfect cell lines (e.g., Sf9, U-2 OS, MEF, MCF7, Hep G2, PC-3, HeLa, HT-29, HT-1080, K-562, RAW 264.7, U-937, Jurkat, PC-12, and HCT 116).
X-tremeGENE HP DNA Transfection Reagent can be successfully used in a variety of applications, such as gene expression analysis and protein production using transiently transfected cells, generation of stable cell lines, expression of shRNA for gene knockdown studies, drug discovery programs, and target evaluation. Contents
Supplied in 80% ethanol and sterile-filtered through 0.2 µm pore size membrane. Does not contain any ingredients of human or animal origin.
Typical Experiment
 High level reproducible transfection efficiency of the X-tremeGENE HP DNA Transfection Reagent. Four hard-to-transfect cell lines were transfected in serum-containing medium using the new X-tremeGENE HP DNA Transfection Reagent. The X-tremeGENE HP DNA Transfection Reagent consistently outperformed the competitor reagent LTX. Transfection efficiency was measured as a ratio of GFP-transfected cells versus whole cell number using the Roche Cellavista Cell Analyzer System; error barrs show the standard deviation of the mean of triplicates.

Widely used adherent HEK-293 and HeLa cells, as well as K-562 cells in suspension, were efficiently transfected in serum-containing medium using the new X-tremeGENE HP DNA Transfection Reagent; the reagent-to-microgram DNA ratios indicated were used. All cell types showed the highest levels of GFP expression using X-tremeGENE HP DNA Transfection Reagent.

Quality
Each lot of X-tremeGENE HP DNA Transfection Reagent is carefully tested following established quality procedures to assure that the product is consistently performing according to specifications.
Cells are transfected with a reporter gene vector DNA using X-tremeGENE HP DNA Transfection Reagent (ratio 3:1 μl/μg DNA). Reporter gene activity is monitored via chemiluminescent detection. Using a standard curve, the total amount of recombinant protein is determined per well in order to meet specification.
