用途:地西他濱作為特異的 DNA甲基化轉(zhuǎn)移酶抑制劑 ,可逆轉(zhuǎn) DNA的甲基化過(guò)程,誘導(dǎo)腫瘤細(xì)胞向正常細(xì)胞分化或誘導(dǎo)腫瘤細(xì)胞凋亡。在腫瘤細(xì)胞內(nèi),地西他濱被脫氧胞苷激酶磷酸化,以磷酸鹽的形式與 DNA摻合。高濃度的地西他濱摻合可抑制 DNA合成誘導(dǎo)細(xì)胞死亡,發(fā)揮其細(xì)胞毒作用;低濃度的地西他濱摻合可替代腫瘤細(xì)胞內(nèi)的胞嘧啶與 DNA甲基化轉(zhuǎn)移酶共價(jià)結(jié)合,使 DNA甲基化轉(zhuǎn)移酶失活但不會(huì)導(dǎo)致細(xì)胞死亡。

General description Decitabine is an epigenetic modifier that inhibits DNA methyltransferase activity which results in DNA demethylation (hypomethylation) and gene activation by remodeling “opening” chromatin. Genes are synergistically reactivated when demethylation is combined with histone hyperacetylation.
