鏈霉蛋白酶消化酶的配制
貯存液濃度20mg/m1,-20℃保存。最佳反應緩沖液為0.01mol/LTris—HCl(pH7.8),0.01mol/LEDTA和0.5%SDS。常用工作濃度lmg/m1,工作溫度37℃。
消化酶的選擇
在選擇用何種酶消化時,應針對要顯示的抗原成分不同而選用不同的酶。一般胃蛋白酶和菠蘿蛋白酶消化主要用于細胞間質(zhì)抗原的檢測,例如纖連蛋白(fibronectin)、層黏連蛋白(1aminin,LN)和各型膠原的顯示;其余酶消化均可用于細胞內(nèi)抗原的檢測。在配制蛋白酶時,應考慮到酶的最佳pH。例如胰蛋白酶的最佳pH為7.6,要在鈣離子的激活下于37℃才能達到最佳效率;而胃蛋白酶則在酸性pH值下才能達到最佳效率。另外,經(jīng)酶消化后的切片應充分洗滌,以去除殘留的消化酶,否則會對抗體或組織進行緩慢消化,影響抗原抗體的結(jié)合,破壞組織結(jié)構(gòu)。陽性和陰性對照片應同時消化,避免人為差異。
